The gain-of-function mutation Card14DE138 has been associated with severe psoriasis and pityriasis rubra pilaris in humans. Mellett and colleagues tested its pathogenicity by introducing Card14DE138 into mouse germline DNA via CRISPR technology. Murine “psoriasis” that shared features with human psoriasis resulted.
