Functional correction of COL7 and anchoring fibril formation at the dermal-epidermal junction.

A recurrent frameshift mutation inCOL7A1 was corrected in primary human RDEB fibroblasts using CRISPR/Cas9 based methodology. Injection of gene corrected fibroblasts into murine COL7 knockout mouse skin that had been grafted onto immunocompromised mice reduced epidermal-dermal cleft formation (top panels) inconjunction with deposition of human COL7 near the basement membrane zone ascompared to uninjected skin (remaining panels).
